naica® Droplet Chip Digital PCR System Accurately Evaluates Korean Beef Molecular Markers to Assist Germplasm Identification

Bos taurus coreanae is a domesticated mammal that serves as a food resource in the Korean consumer market. Its beef consumption far exceeds that of other breeds, and this consumption pattern has led to the emergence of molecular research that distinguishes Bos taurus from other cattle breeds. Not only cattle, but also different breeds of economic animals have significant differences in their economic value in the market, so accurate germplasm identification is imperative.

In a previous study, traditional PCR methods and Sanger sequencing were used to validate the identification of Korean cattle specific SVs generated by TE associated deletion events. It can be used as a molecular marker to distinguish between different cattle breeds (i.e. Korean cattle and Holstein cattle). However, PCR has defects and each sample has various final copy quantifications. In order to overcome the limitations of traditional PCR and accurately evaluate the Korean cattle specific SV sites identified in previous studies, the Department of Biomedical Sciences at Tanguo University, in collaboration with the Livestock Research Institute and Tanguo University School of Medicine, used naica® The droplet chip digital PCR system has achieved more accurate detection of specific SV sites in Korean cattle, and the results have been published in the Journal of Genomics&Informatics as a quantitative evaluation of the molecular marker using droplet digital PCR.

Transposition elements (TEs) account for approximately half of the bovine genome. They can be a powerful species-specific marker without structural variation (SV) regression mutations during genome evolution.
